Replicative or mitotic mutants are an extremely powerful tool for understanding the molecular basis of cell division in eukaryotes. Replicative and mitotic mutants in the budding yeast, S. cerevisiae, have been identified for the rRNA transcription and processing genes (rho). The rho mutants are impaired at the nonsense codon tRNA processing step for rRNA precursors. The rho mutants express full length tRNAs, the mature tRNA species, and precursor tRNAs corresponding to the AUG start codon in the rRNA gene. The levels of these species, however, are reduced by about 100-fold. The reduction in tRNA is due to a translational defect. Initiation at the rho nonsense codon is relatively inefficient. The initiation factor eIF-4GI is required for most efficient initiation in vitro. Mutations at 5 of the 26 rho tRNAs result in a phenotype that is phenotypically more severe than that of mutations in the 3-4 mature rRNA species. The difference in severity of the phenotypes between the rho and 5 mutants suggests that each of the rho mutants may affect initiation at the rho tRNA in a different way. The mutant alleles rho-11 and rho-12 of eIF-4G1 have been mapped and the rho-12 gene has been disrupted. The rho-12 mutants have a slower growth rate than the rho-11 mutants; however, they are blocked at a later step in translation than the rho-11 mutants. Transcription of the rho-12 mutant alleles from a GAL1p-regulated promoter is inhibited in the rho-12 mutant cells, but is unaltered in rho-11 mutant cells. These studies define a role for eIF-4GI in rho nonsense codon suppression and show that eIF-4GI activity is required for most efficient translation of rho mRNA. Cough is one of the most important defensive reflexes of the airway. The final common pathway of the central mediation of cough is a group of limbic, paralimbic, and brainstem nuclei that form a part of the rostral ventrolateral medulla (RVLM). Cough-sensitive (CVLM) neurons are concentrated in the rostral ventrolateral medulla, although the precise anatomical subdivision of the CVLM is debated. In anesthetized cats, pontine stimulation elicited a combination of cough and elevated intra-thoracic pressure (P(IC) and usually P(EB) were present. After extensive transection of the pontine fibers projecting to the CVLM, P(IC) and P(EB) could still be evoked. The transsection of the fibers impinging on the CVLM could therefore be the main cause of the absence of the cough but left intact the vasoactive and respiratory effects of the pontine nuclei. Increasing the frequency of pontine stimulation (theta-rhythm; 3-8 Hz) evoked larger P(IC) and P(EB) but this effect was absent in pontine-cervical (P-C) double-transected animals.
